Application of both LC-MS and LC-NMR to a partially purified ext. of Vernonia fastigiata led to the direct identification of antibacterial sesquiterpene lactones I (R = .beta.-H, R1 = R3 = H, R2 = methacroyl; R = .beta.-H, R1 = R3 = H, R2 = isobutyroyl; R = .beta.-H, R1 = H, R2 = methacroyl, R3 = acetyl; R = .beta.-H, R1 = H, R2 =isobutyroyl, R3 = acetyl; R = .beta.-H, R1 = H, R2 =angeloyl, R3 = acetyl; R = .beta.-H, R1 = R3 = acetyl, R2 = methacroyl; R = .alpha.-H, R1 = H, R2 = methacroyl, R3 = acetyl; R = .alpha.-H, R1= H, R2 =isobutyroyl, R3 = acetyl) and II (R1 = H, R2= methacroyl, R3= acetyl) without isolation of individual compds. The rapid structural anal. of both major and minor components of this class of compds. demonstrated the power of structure-guided screening as a complementary method to assay-guided screening.
